Resumen

AISLAMIENTO Y CARACTERIZACIÓN DE CÉLULAS MADRE ADULTAS PLURIPOTENCIALES DE MEDULA OSEA HUMANA Y DE MONO CYNOMOLOGUS MEDULA OSEA HUMANA Y DE MONO CYNOMOLOGUS #RESUMEN: mapcs (multipotent adult progenitor cells) have been recently described as a population of adult pluripotent stem cells. We have successfully isolated and characterized a cell population from Bone-Marrow of adult human (hMAPC) and adult cynomologus monkey (pMAPC), that co-purify with mesenchymal stem cells. pMAPC proliferate over 150 PD and hMAPC reached 60 PD, both without apparent senescence. h/pMAPC express stem cells markers (Oct4, SSEA4 and Nanog) and lack expression of tissue stem cells specific antigens (CD44, Class I and II, CD31,CD34,CD45), demonstrating a phenotype consistent with human MAPC described previously by Dr. Verfaillie group. Within h/pMAPC populations, we can positively identify a "small population of cells carrying high levies of self-renewal stem cell markers. In vitro, h/pMAPC differentiates into cells expressing markers of mesodermal derived tissues (bone, cartilage, muscle, endothelium) cells, neuroectoderm (neurons) and endoderm (hepatocytes). Also, no transformation or cytogenetic abnormalities has been observed. Interestingly, these populations of h/pMAPC can be isolated and identified without prolonged culture (as early as after 10-20 PD) allowing early expansion of cells. This cell population may have a great clinical and pre-clinical potential in cell therapy.
